This project uses a novel genetic marker - single nucleotide polymorphism (SNP) site analysis, which can provide a complete solution for individual SNP detection. Including sample nucleic acid extraction, second generation sequencing library construction, individual SNP-specific site targeting and individual SNP personalized analysis. Compared with the traditional capillary electrophoresis platform, the high-throughput sequencing method can detect the mixed spots more effectively and quantitatively analyze the contributors of the mixed spots. The success rate of DNA typing was higher for degraded samples such as trace free DNA and severe fragmentation. SNP haplotype analysis has the characteristics of high polymorphism and low mutation rate, and can provide more information than short tandem repeats (STR) alone. Therefore, individual SNP detection has a wider application in forensic individual identification, kinship determination, family origin inference, phenotypic prediction, etc.
