1. The data of different cross multiplicities showed excellent performance

Figure 1. Performance of QKGEN® common DNA hybridization capture kit on different hybridization multiplicities. Multiple hybridization captures of 8plex and 16-plex at 250ng/ library input were performed using the QKGEN® common DNA hybridization capture kit and a 2Mb T Company customer panel, and sequenced using Illumina NoveSeqTM PE150, respectively.

2. High coverage

Figure 2. Performance of QKGEN® common DNA hybridization capture kit in coverage. A 200ng/ library input 8plex multiple hybridization capture of different panels was performed using the QKGEN® common DNA hybridization capture kit and sequenced using Illumina NoveSeqTM PE150. The results showed that the QKGEN® common DNA hybridization capture kit had high coverage for 300Kb, 1Mb, 2Mb, and 34Mb panels.

3. Excellent uniformity

Figure 3. Performance of the QKGEN® common DNA hybridization capture kit on uniformity. A 200ng/ library input 8plex multiple hybridization capture of different panels was performed using the QKGEN® common DNA hybridization capture kit and sequenced using Illumina NoveSeqTM PE150. The Fold-80 base penalty is the additional sequencing multiple required to increase 80% of the bases to average coverage. Lower penalty points indicate that the coverage depth of different target areas is less different. In the case of perfect unity, the penalty is 1. The results showed that the QKGEN® common DNA hybridization capture kit demonstrated excellent uniformity for 300Kb, 1Mb, 2Mb, and 34Mb panels, with a Fold-80 base penalty of less than 1.5.

4. Excellent capture efficiency

The capture efficiency refers to the proportion of sequences (effective sequences) that are compared to the target region in all sequences compared to the reference genome. The capture efficiency does not affect the data quality, but only the effective proportion of the data. The more efficient the capture, the smaller the amount of data needed to cover the same depth, and the lower the sequencing cost.

Figure 4. Performance of QKGEN® common DNA hybridization capture kit on target rate. A 200ng/ library input 8plex multiple hybridization capture of different panels was performed using the QKGEN® common DNA hybridization capture kit and sequenced using Illumina NoveSeqTM PE150. The results showed that the QKGEN® common DNA hybridization capture kit demonstrated excellent capture efficiency and low repetition rates for 300Kb, 1Mb, 2Mb, and 34Mb panels.
